Human platelet lysate improves the growth and survival of cultured human pre-antral follicles

Research questionHow does platelet rich plasma products like human Platelet Lysate (hPL) and umbilical-cord blood (UCP) affect the growth survival of isolated pre-antral follicles in vitro?DesignHuman (n=724; mean diameter: 75 µm; range: 46-237 µm) were from ovarian medulla donated by 14 patients undergoing unilateral oophorectomy for tissue cryopreservation. Follicles encapsulated 0.5% alginate cultured 8 days media supplemented with: 5% Fetal Bovine Serum (FBS, n=171), 2.5% Human Albumin (HSA, n=159), hPL (n=223), or UCP (n=171).ResultsThe probability was significantly higher group with (80%) compared to other three groups: FBS (54%); HSA (63%) (29%). Surviving had less defined follicular membranes decompacted granulosa cell layers. The average surviving larger (73 any (43 μm); (40 μm) (54 μm). A descriptive analysis secretion AMH oestradiol did not reveal difference between groups. detectability genes high AR (100%), AMHR2 FSHR (76%), whereas few expressed LHR (20%).ConclusionHuman lysate improved FBS, UCP. use is a valuable improvement culture but further studies will have prove whether superiority translates into better quality oocytes.
